Description
These data are the result from the final stage (Manaus, Brazil) of the XPRIZE Rainforest competition. During 24 hours data was captured (predominantly audio and eDNA) and it was analyzed during the 48 hours afterwards. The data contains the unique species that were detected at each location through both methods.

Données sur le projet
The XPRIZE Rainforest competition is a contest where teams have 24 hours to obtain samples from a 100 ha area, without being able to physically enter the zone. The data was collected by monitoring platforms deployed by remotely operated drones during the first 24 hours of the competition. After the platforms were retrieved the data was analyzed in the subsequent 48 hours to report on the biodiversity and general health of the monitored area.

Description du design | The objectives during the 24 hours of data collection were to collect audio recordings and organic material for eDNA analysis from 6 monitoring platforms installed in the tree canopy distributed over the competition area. Separate drone flights were organized to collect additional organic material from various spots in the competition area. |

Méthodes d'échantillonnage
Six monitoring stations (DROPs) were deployed covering the 100 ha competition area. Each DROP included a digital microphone sampling continuously at 192 kHz and 16 bits. The acoustic data was preprocessed on the DROP itself (sound level measurements, ecoacoustic indicators, acoustic classification), and processed further after recovery. The DROP also has a few "sticky" zones where organic material (mostly insects) can be collected. The systems were left in the trees during the night and recovered the following morning; all systems needed to be back in the base camp before noon local time.

Description des étapes de la méthode:
- The acoustic data was processed by custom software written in C or Python. The classification was done through a CNN that was trained on data from various sources. In addition, the audio was processed by BirdNET. The GBIF dataset consists of results from both classifiers where we included the best example from each detected species at each DROP. The eDNA analysis utilizes universal primers to detect plants and animals in one single assay for a comprehensive species identification. The experimental workflow allowed for high sample throughput at high quality. This was achieved through a universal, field-ready DNA extraction protocol, pooled library preparation, efficient library normalization strategy and real time analyses and species identification during sequencing. The protocol is suitable for metabarcoding and barcoding, as well as long-range PCR. The implementation of long-range PCR has the potential to inform about population individual composition enabled by Nanopore's unique long-read capabilities.
